Ran-binding protein 6 (RANBP6) is a member of the importin β family of nuclear transport receptors, predicted to facilitate the import of proteins bearing nuclear localization signals into the nucleus[2][3]. RANBP6 interacts with Ran-GTP and related transport proteins and contains HEAT repeats and a putative Ran-binding domain[1]. It forms part of the machinery regulating the nuclear import of specific cargoes, including transcription factors such as STAT3[1]. Recent studies indicate RANBP6 can repress the transcription of EGFR by promoting STAT3 nuclear translocation and binding to the EGFR promoter, constituting an indirect feedback mechanism in EGFR pathway regulation. Disruption or silencing of RANBP6—such as by genetic deletions—has been implicated in increased EGFR expression and signaling in glioblastoma, suggesting a functional role in cancer biology, though it is not currently a direct drug target[1][8].
