Raf refers to a family of serine/threonine-specific protein kinases that play a central role in cell signaling, particularly in the RAS-RAF-MEK-ERK (MAPK) pathway. There are three main RAF kinase isoforms in humans: A-RAF, B-RAF, and C-RAF (also known as Raf-1). The primary function of RAF kinases is to transmit signals from activated RAS proteins at the cell membrane to downstream effectors by phosphorylating MEK1/2, which then activate ERK1/2. Mutations leading to aberrant activation of RAF proteins are implicated in cancer development, particularly the B‑RAFV600E mutation.
