The Ras–Raf interaction is a critical molecular event in the MAPK/ERK signaling pathway, regulating essential cellular processes. It involves direct binding between activated (GTP-bound) Ras proteins and Raf kinases (A-Raf, B-Raf, C-Raf/Raf-1). This interaction recruits Raf to the plasma membrane, relieving autoinhibition and allowing activation of the MAPK/ERK cascade. Aberrant activation, often due to mutations in RAS or RAF, is implicated in many cancers, making this interaction a therapeutic target.
