Overview

Ras and Rab interactor 1 (RIN1) is a multifunctional intracellular effector protein that interacts with activated Ras and Rab proteins. It modulates signal transduction pathways by acting as a Ras effector, a guanine nucleotide exchange factor (GEF) for Rab5, and an activator of ABL family tyrosine kinases[1][4]. RIN1 competes with RAF1 for binding to activated Ras, enhancing signaling from ABL kinases (involved in cytoskeletal remodeling) and facilitating receptor endocytosis, particularly for the EphA4 receptor in neurons[1][3]. It is involved in neuronal plasticity, memory formation, and synaptic strength regulation, as well as intracellular trafficking via Rab5. RIN1 is associated with diseases such as bone disease and colorectal cancer, but is not a direct drug target. Research drugs, such as Dasatinib (a broad-spectrum tyrosine kinase inhibitor), have been used in pathway studies involving RIN1, particularly regarding neuronal signal modulation and endocytosis[3][1].\n\nNote: RIN1 is not a conventional therapeutic target (such as a receptor, enzyme, or transporter typically targeted by drugs), but rather a signaling adaptor and regulatory protein that modulates pathways connected to Ras and Rab small GTPases[1][2].
