Overview

Ras and Rab interactor 3 (RIN3) is a multifunctional protein and a member of the RIN family, acting as a guanine nucleotide exchange factor (GEF) for Rab5 and Rab31 GTPases, and participating in intracellular vesicular trafficking, particularly in early endocytosis[1][2][3]. It contains SH2, proline-rich, RH, VPS9, and Ras association domains, allowing interactions with Ras GTPases and receptor tyrosine kinases; its activity is modulated by tyrosine phosphorylation[1][2]. RIN3 influences various cell signaling pathways through its regulatory roles in endocytosis, trafficking of membrane proteins such as transferrin and mannose 6-phosphate receptor, and mediates interactions with partners such as CD2-associated protein (CD2AP) and BIN1[1][2][3]. Genetic variants in RIN3 are implicated in diseases including Alzheimer's disease, Paget’s disease of bone, chronic obstructive pulmonary disease, and obesity; these may affect the risk and pathogenesis via altered expression or missense mutations in functional domains[1][2]. While not currently a direct therapeutic target, it is under study for its relevance as a disease biomarker and as a potential mediator of disease mechanisms, especially in neurodegeneration and bone homeostasis[1][2].
