Overview

Ras association domain-containing protein 2 (RASSF2) is a tumor suppressor protein belonging to the Ras association domain family, characterized by non-enzymatic adaptor functions. RASSF2 contains a Ras association domain and a C-terminal SARAH (Salvador-Rassf-Hippo) domain, and serves as a KRAS-specific effector protein. Its principal roles include modulation of Ras and Hippo signaling, stabilization of Hippo kinases MST1 and MST2, promotion of apoptosis, and cell cycle arrest. RASSF2 suppresses tumorigenesis and is often transcriptionally silenced in cancer via promoter hypermethylation. It acts as a critical inhibitor in leukemia models, notably acute myeloid leukemia with t(8;21) translocation, and influences Rac GTPase signaling. Restoring RASSF2 function is a research focus in cancer therapeutics due to its involvement in multiple tumor-suppressive pathways.
