Overview

Ras association domain family member 3 is a ubiquitously expressed protein containing a Ras association (RA) domain that acts as a Ras effector and a tumor suppressor. RASSF3 directly interacts with the E3 ubiquitin ligase MDM2, facilitating its ubiquitination and stabilization of p53, thus promoting p53-dependent transcription of genes that regulate cell cycle arrest and apoptosis. Loss or silencing of RASSF3 impairs DNA damage-induced apoptosis, disrupts cell-cycle checkpoints, and diminishes p53-mediated genomic integrity. Its function is impaired in certain tumors, notably pituitary adenoma and cancers associated with specific single nucleotide polymorphisms. It is not a receptor, enzyme, transporter, or transcription factor, but rather a signal transduction and tumor suppressor protein with a highly conserved domain structure.
