Ras association domain family member 6 (RASSF6) is a tumor suppressor protein belonging to the RASSF (Ras association domain family) family. It contains Ras Association (RA) and SARAH (Salvador/RASSF/Hippo) domains and uniquely features a PDZ-binding motif. RASSF6 is frequently silenced by epigenetic mechanisms in various human cancers and functions as a regulator of apoptosis and cell cycle arrest. Mechanistically, RASSF6 stabilizes p53 by blocking MDM2-mediated degradation but can also induce cell cycle arrest and apoptosis independently of p53, in part by modulating the retinoblastoma protein (pRb) and E2F1 axis. RASSF6 interacts with components of the Hippo pathway and modulates their activity, linking this pathway to tumor suppression. Loss or downregulation of RASSF6 is associated with poor prognosis and increased tumor aggressiveness, supporting its function as a tumor suppressor[1][2][4].
