Overview

The RAS guanyl-releasing protein family comprises a group of diacylglycerol-regulated guanine nucleotide exchange factors that specifically activate members of the small GTPase superfamily such as Ras and Rap by facilitating GDP/GTP exchange. These proteins play critical roles in signal transduction pathways controlling lymphocyte development, homeostasis, differentiation, and function. Deficiencies or mutations in these proteins can result in primary immunodeficiencies with increased susceptibility to infections and lymphoproliferative diseases. The family includes several members—RAS guanyl-releasing protein 1 (RASGRP1), 2 (RASGRP2), 3 (RASGRP3), and 4 (RASGRP4)—each with tissue-specific expression patterns and physiological roles. For example, RASGRP1 is essential for T-cell receptor signaling; RASGRP2 is enriched in brain tissue but also linked to platelet function; RASGRPs are generally regulated by calcium ions and diacylglycerol binding domains which control their activation state. Therapeutic targeting is complex due to their central role in immune regulation.
