Ras homolog enriched in brain-like protein 1 (RHEBL1) is a member of the Ras superfamily of small GTPases and shares high sequence similarity with Rheb (Ras homolog enriched in brain)[3]. Like Rheb, RHEBL1 contains a C-terminal CaaX motif required for farnesylation and association with cell membranes. RHEBL1 is capable of activating the mTORC1 (mechanistic target of rapamycin complex 1) signaling pathway, stimulating cell growth and proliferation, particularly evident in overexpression systems. In cells lacking Rheb, RHEBL1 can compensate and restore mTORC1 signaling, indicating functional redundancy[3]. The protein cycles between inactive GDP-bound and active GTP-bound states, with GTP-bound RHEBL1 promoting mTORC1 activation. Increased activity or overexpression of RHEBL1 may play roles in diseases associated with dysregulated cell growth, such as cancer; however, it has not been characterized as a direct drug target, and little is known about specific drug interactions or therapeutic safety concerns. RHEBL1 occupies a niche as a signaling node in cellular growth regulation via the mTOR pathway[1][2][3].
