Overview

Ras-related protein Rab-25 (RAB25) is a small GTPase belonging to the Rab subfamily of the RAS superfamily of proteins[4][7]. It is primarily expressed in epithelial cells and regulates the trafficking and recycling of vesicles, particularly those involved in the recycling of cell-surface receptors to the plasma membrane[1][3]. Rab25 modulates key cellular processes such as cell proliferation, motility, survival, and signal transduction. Aberrant expression has been linked to the progression, aggressiveness, and poor prognosis of multiple cancers—including ovarian, breast, renal, gastric, lung, bladder, liver, glioblastoma, and prostate cancers—usually promoting oncogenesis, but it may act as a tumor suppressor in certain contexts such as colorectal and some head and neck cancers[1][2][3]. Mechanistically, Rab25 affects oncogenic pathways (Akt, Wnt, Src) and the recycling/function of integrins and other receptors, and its altered activity can lead to chemoresistance and changes in tumor cell phenotype[3]. No direct pharmacologic inhibitors are currently approved, but Rab25's context-dependent biology makes it an active area of research for therapeutic targeting and biomarker development.
