Ras-related protein Rab-28 (RAB28) is a member of the Rab subfamily of Ras-related small GTPases, which are essential regulators of intracellular membrane trafficking[2][5][7][11]. RAB28 is highly expressed in photoreceptor cells, particularly at the ciliary basal body and rootlet of retinal cells, where it is necessary for the shedding and phagocytosis of cone outer segment discs, a key process for photoreceptor maintenance and vision[2][8][9][3]. Pathogenic variants in RAB28 are causative for rare, autosomal recessive forms of cone-rod dystrophy, a retinal degenerative disorder characterized by photophobia, visual acuity loss, and central retinal thinning[2]. RAB28 itself is not currently recognized as a therapeutic target for pharmacological intervention, and there are no drugs known to specifically target this protein. Despite being enzymatic (as a GTPase), its roles are largely structural and trafficking-related in retina, without direct targeting by available drugs.
