Overview

Ras-related protein Rab-2A (RAB2A) is a member of the Rab family of small GTPases within the Ras superfamily[1][4][8]. These are monomeric, membrane-bound proteins that regulate intracellular membrane trafficking, vesicle formation, transport, tethering, and fusion through their cycle between active (GTP-bound) and inactive (GDP-bound) states[5][8]. RAB2A specifically facilitates the transport of proteins from the endoplasmic reticulum to the Golgi apparatus and is essential for maintaining Golgi morphology and autophagic flux by mediating autophagosome-lysosome fusion[1][8]. RAB2A influences cellular processes including junctional stability, cell compaction, and apoptosis, and has documented interactions in neurodevelopment and tumor invasiveness[3][6]. Variants in RAB2A are associated with some cancers and have reported links to neurodevelopmental disorders like autism[1][2]. The protein operates through recruitment of endosomal tethering complexes and may regulate switching between secretion and protein quality control depending on metabolic stress[9].
