Ras-related protein Rab-30 (RAB30) is a member of the Rab subfamily within the Ras superfamily of small GTPases, which are key regulators of intracellular membrane trafficking[1][2][3]. RAB30 is predominantly localized to the Golgi apparatus, trans-Golgi network (TGN), and recycling endosomes in a variety of cell types, where it mediates GTP binding and GTPase activity[1][2][3]. RAB30 is essential for maintaining the structural and functional integrity of the Golgi apparatus and is involved in retrograde trafficking of cargo proteins from endosomes to the Golgi, particularly in regulating TGN38 transport[1][2]. Emerging evidence indicates that RAB30 plays critical roles in host–pathogen interactions, including Golgi remodeling during Toxoplasma gondii infection and anti-bacterial autophagy against group A Streptococcus[1]. While associated with disorders such as syndromic X-linked intellectual disability and Warburg Micro Syndrome 1, RAB30 is not currently established as a direct therapeutic target[3].
