Ras-related protein Rab-36 (RAB36) is a small GTPase of the Rab family within the Ras superfamily, encoded by the RAB36 gene located on human chromosome 22[3][7]. It functions as a molecular switch, regulating intracellular membrane trafficking by controlling the formation, movement, and fusion of vesicles within cells[6][8]. RAB36 is involved in retrograde transport of melanosomes in melanocytes through its interaction with the Rab-interacting lysosomal protein (RILP) and related effectors, utilizing a RILP homology domain (RHD) to mediate these interactions[2][4]. In neurons, RAB36 is highly expressed and implicated in axon-specific trafficking events, such as the movement of organelles and possibly synaptic components, though the physiological and pathophysiological relevance of this role is still under study[2]. To date, RAB36 is not considered a therapeutic target, and no specific drugs, clinical biomarkers, or known therapeutic liabilities are associated with it[6][8].
