Overview

Ras-related protein Rab-3D (RAB3D) is a member of the Rab family of small GTPases that serves as a key regulator of regulated exocytosis, particularly in exocrine cells like those found in the pancreas and lacrimal glands (UniProt P40306). It facilitates the recruitment of secretory granules to the plasma membrane and their subsequent fusion, a process essential for the secretion of enzymes and other proteins (NCBI Gene: 9545). In the context of pathology, RAB3D mRNA is frequently overexpressed in several types of cancer, where it promotes tumor progression by inducing epithelial-mesenchymal transition (EMT) and enhancing cell migration and invasion (PubMed: 28423615). Research indicates that silencing RAB3D mRNA using siRNA can significantly reduce the metastatic potential of cancer cells, making it a target of interest for RNA-based therapeutics (PubMed: 30605344). While there are currently no FDA-approved drugs targeting RAB3D, its involvement in the secretome of cancer cells and its correlation with poor clinical outcomes highlight its potential as both a biomarker and a therapeutic target.
