Ras-related protein Rab-42 (RAB42) is a small GTPase of the Ras oncogene family, involved primarily in the regulation of intracellular vesicular trafficking between endosomes and the trans-Golgi network[1][4]. RAB42 acts as a negative regulator of TLR9 and TLR4 signaling pathways, affecting immune responses in macrophages, and is implicated in modulating NF-κB/STAT3 signaling in megakaryocytic differentiation[1]. RAB42 is significantly overexpressed in a wide array of tumors, particularly in advanced cancers, and its high expression is associated with reduced immune cell infiltration, immune checkpoint gene expression, chemotherapy resistance, and poor overall survival. In hepatocellular carcinoma and glioma, RAB42 promotes cell proliferation, migration, and invasion[1][3]. It is considered a novel oncogenic driver, prognostic marker, and potential therapeutic target, especially in liver and brain cancers[1][2][3].
