Ras-related protein Rab-5A (RAB5A) is a small GTPase of the Rab family, part of the Ras superfamily, which acts as a molecular switch to regulate endocytic vesicle trafficking, especially the early stages of endosome fusion[1][2][5]. It cycles between inactive (GDP-bound) and active (GTP-bound) states, controlling the recruitment of effector proteins and facilitating membrane fusion involved in nutrient uptake, receptor downregulation, and antigen presentation[2][7]. RAB5A participates in several signaling pathways, including autophagy and the mTOR signaling pathway, and interacts with various regulators and effectors responsible for endosomal dynamics[2][5].
