Ras-related protein Rab-7A (RAB7A) mRNA encodes a key member of the Rab family of small GTPases, which functions as a master regulator of late endocytic trafficking. The protein product is essential for the maturation of late endosomes and their subsequent fusion with lysosomes, as well as for the progression of autophagy and phagocytosis (UniProt, 2024). Mutations in the RAB7A gene are the underlying cause of Charcot-Marie-Tooth disease type 2B (CMT2B), a peripheral neuropathy characterized by axonal degeneration (PubMed, 2021). In oncology, RAB7A expression levels are often altered, influencing the degradation of signaling receptors and contributing to tumor cell migration and invasion (NCBI, 2023). Furthermore, RAB7A is exploited by various viruses, including SARS-CoV-2 and Ebola, to facilitate intracellular transport during the infection cycle (PubMed, 2022). Therapeutic strategies targeting RAB7A mRNA, such as antisense oligonucleotides (ASOs) and siRNA, aim to modulate protein levels to treat CMT2B or inhibit viral replication, though challenges remain regarding the potential disruption of essential cellular degradation pathways (PubMed, 2023).
