Overview

Ras-responsive element-binding protein 1 (RREB1) is an evolutionarily conserved zinc finger transcription factor that acts downstream of the RAS signaling pathway, binding RAS-responsive elements in gene promoters and influencing gene expression through both repression and activation. RREB1 integrates multiple signaling pathways (e.g., EGFR/MAPK, JAK/STAT, TGF-β/SMAD, Notch, Sonic Hedgehog) and is involved in numerous cellular processes, including regulation of cell adhesion, epithelial architecture, cytoskeletal dynamics, proteostasis, and embryonic development. Mutations or dysregulation of RREB1 contribute to cancer progression and metastasis by promoting epithelial-to-mesenchymal transition, and to developmental syndromes such as Noonan-like RASopathies. In neurons, RREB1 is critical for maintaining the microtubule network and proteostasis, and its deficiency leads to degeneration of Purkinje cells and ataxia, highlighting its role in neurodegenerative disease.
