Ras suppressor protein 1 (RSU1) mRNA encodes a highly conserved leucine-rich repeat (LRR) protein that is essential for the regulation of the Ras signal transduction pathway and cell-extracellular matrix (ECM) adhesion. The RSU1 protein localizes to focal adhesions and interacts with the LIM5 domain of PINCH1 to form the ILK-PINCH-Parvin (IPP) complex, which is critical for actin cytoskeleton remodeling and cell spreading. Although originally identified as a suppressor of Ras-dependent oncogenic transformation, RSU1 exhibits a complex role in cancer, acting as either a tumor suppressor or a promoter of metastasis depending on the cellular context and the specific isoform expressed. Targeting RSU1 mRNA through RNA interference (RNAi) or microRNAs (miRs), such as miR-182-5p and miR-409-3p, has been shown to effectively inhibit the migration and invasion of aggressive cancer cells in vitro. These therapeutic strategies aim to modulate downstream signaling pathways, including PTEN/PI3K/AKT and p38 MAPK, to suppress tumor progression and metastasis. Currently, RSU1 mRNA is primarily a research target, and there are no approved drugs specifically targeting this molecule in clinical use.
