RASSF1 antisense RNA 1 (RASSF1-AS1) is a long non-coding RNA transcribed from the opposite strand of the RASSF1 gene locus at 3p21.3. It functions primarily by forming antisense RNA interactions with the RASSF1A mRNA, thereby suppressing its translation and promoting gene silencing at the epigenetic level by recruiting PRC2 complex component SUZ12, which locally increases H3K27me3 repressive histone marks at the RASSF1A promoter[5][1]. RASSF1-AS1 is upregulated in disease states such as cardiac fibrosis, where it promotes pathological remodeling by inhibiting the tumor suppressor activity of RASSF1A[1]. Its expression is also higher in triple negative breast cancer samples, marking it as a potential cancer biomarker[5]. RASSF1-AS1 is localized in the nucleus and is usually unspliced and polyadenylated[5]. Currently, there are no known drugs directly targeting RASSF1-AS1, and its mechanistic functions relate to gene regulation rather than acting as a classic therapeutic target like a receptor or enzyme.
