Overview

RB-associated KRAB zinc finger protein (RBAK) is a member of the KRAB-containing zinc finger transcription factor family, characterized by an N-terminal KRAB (Krüppel-associated box) domain functioning as a strong transcriptional repressor and a C2H2 zinc finger domain that mediates sequence-specific DNA binding[1][2][3]. KRAB zinc finger proteins are vertebrate-specific, modular proteins that frequently modulate gene transcription by recruitment of corepressors and may participate in diverse biological processes including cell differentiation, cell proliferation, apoptosis, nucleolar maintenance, and cell cycle arrest[1][2]. The RBAK gene product is widely expressed and may interact with the retinoblastoma protein (RB), a well-known tumor suppressor, linking it to potential roles in cell cycle regulation and tumorigenesis[1][3]. No drugs are currently known to target RBAK directly, and its utility as a therapeutic target, biomarker, or safety concern in human therapy has not been established.
