RBM15 antisense RNA 1 (RBM15-AS1) is a long non-coding RNA transcribed antisense from the RBM15 gene locus on human chromosome 1. RBM15-AS1 enhances RBM15 protein translation via overlap with the RBM15 5′UTR, functioning in a CAP-dependent manner. Its upregulation promotes terminal differentiation of megakaryocytes, while its knockdown attenuates this process. RBM15-AS1 and its sense counterpart, RBM15, are both transcriptionally regulated by RUNX1 (a key hematopoietic transcription factor) and are repressed by the RUNX1-ETO fusion protein, common in certain leukemias. While RBM15-AS1 is primarily nuclear, its cytoplasmic levels increase during cellular differentiation signals. As a lncRNA, it provides a layer of post-transcriptional regulation fine-tuning RBM15 protein output, which is critical for proper hematopoietic cell differentiation and may be linked to the pathogenesis of specific leukemias[1][2].
