RBM5 antisense RNA 1 (RBM5-AS1), also known as LUST (LUCA-15-specific transcript), is a long non-coding RNA transcribed antisense to the RBM5 gene. It is predominantly localized in the nucleus and acts as a chromatin-associated regulatory RNA. RBM5-AS1/LUST is involved in the regulation of WNT/β-catenin signaling through direct interaction with β-catenin, promoting assembly of the β-catenin–TCF4 complex at WNT target gene promoters. This lncRNA is critical for maintaining stem-like characteristics and self-renewal of cancer-initiating cells in colon and breast cancer. Its expression correlates with tumor progression, maintenance of stemness, increased cell proliferation, resistance to differentiation, and poorer clinical outcome[1][2][4][5].
Regulates WNT/β-catenin signaling by physically interacting with β-catenin and facilitating the β-catenin-TCF4 transcriptional complex, modulates transcription of WNT target genes (e.g., MYC, CCND1, YAP1, SGK1)[1][2][4][5]
