Overview

RCSD domain containing 1 (RCSD1), also called CapZIP, is an actin-binding protein involved in regulation of the cytoskeleton in immune and muscle cells, linking stress signaling to cytoskeletal remodeling[1]. RCSD1 specifically interacts with actin capping protein CapZ, modulating filament assembly; upon cell stress, RCSD1 undergoes phosphorylation by stress-activated kinases (including MAPKAP-K2 and MAPKAP-K3), causing dissociation from CapZ and remodeling of actin networks[1][2][3]. Genetic alterations such as RCSD1-ABL1 fusion drive certain leukemia subtypes and serve as a biomarker and potential therapeutic target by kinase inhibition[4]. Though RCSD1 is important in cellular structure and immune function, it is not a canonical therapeutic target (receptor, enzyme, ion channel, transporter)[3]; its disease relevance arises mainly through fusion events and altered cytoskeletal regulation[2][4].
