Reactive carbonyl species (RCS) are a class of highly reactive molecules characterized by the presence of one or more carbonyl groups (C=O). These compounds are generated endogenously as metabolic byproducts, primarily through lipid peroxidation and sugar metabolism. Key examples include acrolein, 4-hydroxy-(E)-2-nonenal (4-HNE), and methylglyoxal. RCS can modify proteins, nucleic acids, and lipids, leading to both cellular damage and signaling. While excessive RCS accumulation is linked to pathologies like diabetes complications and neurodegenerative diseases, they also play a role in redox signaling pathways, modulating transcription factors such as Nrf2. Due to their reactivity, targeting RCS presents significant challenges related to specificity and off-target effects.
