Overview

Reactive oxygen species (ROS) and lipid peroxidation products (LPPs) are key mediators of oxidative stress in ocular tissues, particularly the retina and lens (PMID: 23122729). ROS, including superoxide and hydroxyl radicals, are generated as byproducts of mitochondrial respiration and photo-oxidation, while LPPs like malondialdehyde (MDA) and 4-hydroxynonenal (4-HNE) result from the oxidative degradation of polyunsaturated fatty acids (PMID: 21840302). In the eye, these molecules contribute to the pathogenesis of age-related macular degeneration (AMD), cataracts, and diabetic retinopathy by inducing protein carbonylation, DNA damage, and apoptosis (PMID: 25824066). Therapeutic strategies often involve the use of exogenous antioxidants, such as those found in the AREDS2 formulation (Vitamin C, E, Lutein, Zeaxanthin), to scavenge ROS and limit the propagation of lipid peroxidation (NIH/NEI). Although they are not specific protein targets, monitoring their levels serves as a critical biomarker for oxidative damage and therapeutic efficacy in ophthalmic research.
