Overview

Receptor Activator of Nuclear Factor κB Ligand (RANKL) is a type II transmembrane protein belonging to the TNF superfamily. It is primarily produced by osteoblasts and cells of the immune system. RANKL is essential for the formation, function, and survival of osteoclasts, the cells responsible for bone resorption. By binding to its receptor RANK on osteoclast precursors and mature osteoclasts, RANKL stimulates their differentiation and activity. The balance between RANKL and its decoy receptor OPG (osteoprotegerin) is crucial for maintaining bone homeostasis. Dysregulation of the RANKL pathway can lead to various bone diseases characterized by excessive bone loss.
