Overview

Receptor activity-modifying protein 1 (RAMP1) is a single-transmembrane-domain accessory protein that modulates the function, trafficking, and ligand specificity of certain G protein-coupled receptors (GPCRs), most notably the calcitonin receptor-like receptor (CALCRL). When associated with CALCRL, RAMP1 enables the formation of a functional receptor for calcitonin gene-related peptide (CGRP). RAMP1 plays roles in angiogenesis, lymphangiogenesis, and wound healing and represents a targetable interface for selective drugs affecting neuropeptide signaling.

Mechanism of action

Modulation of GPCR function and trafficking through direct interaction. Acts as a chaperone to get CALCRL to the cell surface and alters ligand binding properties of receptors. Targeting RAMP1, or the interface of RAMP1 and its GPCR partners, disrupts downstream signaling pathways, such as those mediated by CGRP.
