Receptor Activity-Modifying Protein 2

Molecular classification
Accessory membrane protein, Chaperone protein, GPCR modulator
01

Overview

Receptor activity-modifying protein 2 (RAMP2) is an accessory membrane protein that interacts with and modulates the function of several G-protein coupled receptors (GPCRs), particularly those in the class B family. RAMP2 is essential for determining receptor pharmacology, trafficking, and signaling specificity. It primarily functions by allosterically binding to certain GPCRs, such as the glucagon receptor (GCGR) or the calcitonin receptor-like receptor (CALCRL), influencing their trafficking, localization, and signaling properties. It plays a critical role in cardiovascular physiology, and its loss-of-function can result in severe defects.

Mechanism of action

Modulates ligand selectivity, regulates endocytic trafficking, and alters intracellular retention and recycling dynamics of target GPCRs, influencing their signaling bias.
