Overview

Receptor activity-modifying protein 3 (RAMP3) is a single-transmembrane-domain protein that belongs to the RAMP family. It modulates the function and trafficking of certain G protein-coupled receptors (GPCRs), especially those in the calcitonin receptor family. RAMP3 is required for transporting specific GPCRs, such as calcitonin receptor-like receptor (CALCRL) and G-protein coupled estrogen receptor (GPER1), from the endoplasmic reticulum/Golgi to the plasma membrane. When associated with CALCRL, it forms the adrenomedullin receptor AM2. When associated with Calcitonin receptors, forms amylin receptor AMY3. It plays a role in various signaling pathways and cardioprotection.

Mechanism of action

RAMP3 modulates the function and trafficking of certain G protein-coupled receptors (GPCRs), especially those in the calcitonin receptor family. It forms complexes with CALCRL and Calcitonin receptors resulting in receptors with altered ligand specificity. When associated with CALCRL: - With RAMP3 → forms dual CGRP/AM receptor AM2, but primarily acts as an adrenomedullin receptor with low affinity for CGRP[2][5]. When associated with calcitonin receptors: - Forms amylin receptors; specifically, CALCR + RAMP3 = amylin receptor AMY3[2].
