Receptor-interacting protein kinase 2 (RIPK2) is a serine/threonine-protein kinase essential for both innate and adaptive immune responses. It acts as a key downstream signaling molecule for NOD-like receptors, specifically NOD1 and NOD2, mediating pro-inflammatory signaling by activating NF-κB and MAPK pathways. RIPK2 is involved in cell death regulation, promoting apoptosis and potentially necroptosis. It also promotes autophagy and is phosphorylated upon DNA damage. RIPK2 plays a central role in chronic inflammatory conditions and cancer progression, making it a validated target for small molecules and PROTACs.
