Receptor-operated calcium channels are a class of ion channels permeable to calcium ions, activated in response to agonist binding at G-protein-coupled receptors or other cell surface receptors, independent of membrane depolarization. They often consist of transient receptor potential canonical (TRPC) channel subunits, and are found in various tissues, especially smooth muscle and vasculature, where they mediate calcium influx in response to stimuli like neurotransmitters and hormones. ROCCs regulate key functions including smooth muscle contraction, vascular tone, and cell proliferation, and are implicated in diseases such as hypertension, vascular remodeling, and some cancers. Currently, there are no highly specific drugs targeting ROCCs in clinical use, though experimental compounds exist. Their close relationship with store-operated calcium channels (SOCCs) and molecular overlap with other TRP channels present challenges for therapeutic targeting.
Inhibition or modulation of receptor-operated calcium channels diminishes receptor-induced calcium influx, which can reduce smooth muscle contraction, cell proliferation, or other downstream signaling. Often acts through blockade of TRP channels (primarily TRPC subtypes) or through modulation of the PI(4,5)P2/phospholipase C pathway.
