Overview

Receptor protein-tyrosine kinase erbB-3 (ERBB3/HER3) is a member of the epidermal growth factor receptor (EGFR/ERBB) family. It is a receptor tyrosine kinase that, upon activation by neuregulins, primarily heterodimerizes with other ErbB receptors, most notably ErbB2/HER2, to activate downstream signaling pathways, particularly the PI3K/Akt pathway. While it possesses impaired kinase activity, it plays a crucial role in cell survival, proliferation, and drug resistance in various cancers, making it an important therapeutic target.
