Receptor-transporting protein 1 (RTP1) is a membrane accessory protein primarily expressed in the olfactory epithelium, where it facilitates the trafficking and cell-surface expression of olfactory (odorant) receptors, a subset of G-protein-coupled receptors (GPCRs).[1][3][5] RTP1 interacts directly with odorant receptors, enabling their exit from the endoplasmic reticulum and subsequent localization to the plasma membrane, a process not typically achieved in heterologous cells absent RTP1. RTP1 does not promote expression of non-olfactory GPCRs, indicating functional specificity.[3][5] The gene encoding this protein is also referred to as Z3CXXC1 and possesses a zinc finger domain. There is no established evidence that RTP1 is a therapeutic drug target, nor are there known drugs, biomarkers, or safety issues specifically associated with it. Its primary biological role is supporting the sense of smell through odorant receptor maturation and trafficking[1][3][5].
