ROR1 mRNA is the messenger RNA transcript that encodes the Receptor tyrosine kinase-like orphan receptor 1, a type I transmembrane protein and oncofetal antigen. While the ROR1 protein is a well-established therapeutic target due to its high expression in various hematological and solid malignancies and its absence in most adult tissues, the mRNA itself serves as a critical target for nucleic acid-based therapies such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs). Targeting ROR1 at the mRNA level aims to silence the expression of the protein, thereby disrupting non-canonical Wnt5a signaling pathways that drive tumor cell survival, epithelial-mesenchymal transition (EMT), and metastasis. In clinical and research settings, ROR1 mRNA levels are frequently utilized as a biomarker for disease progression and as a prognostic indicator, particularly in chronic lymphocytic leukemia (CLL) and non-small cell lung cancer (NSCLC).
