Overview

Receptor tyrosine-protein kinase erbB4 (HER4) is a member of the ErbB family of receptor tyrosine kinases. It plays critical roles in the development and maintenance of the cardiovascular system, central nervous system, and mammary gland. Ligand binding, such as Neuregulins, promotes dimerization and activation of downstream signaling pathways like MAPK/ERK. Mutations in ERBB4 have been identified in various cancers and neurodegenerative diseases. Multiple isoforms exist due to alternative splicing, leading to functional diversity.
