The red blood cell (RBC) membrane is a specialized structure composed of a lipid bilayer tethered to a complex protein cytoskeleton, primarily consisting of spectrin, actin, and protein 4.1 (Mohandas & Gallagher, 2008). This architecture provides the erythrocyte with the remarkable durability and deformability required to survive 120 days in circulation and pass through capillaries smaller than its own diameter (Lux, 2016). Key integral proteins include Band 3 (anion exchanger 1), which facilitates CO2 transport, and glycophorins, which contribute to the membrane's negative charge and serve as receptors for pathogens (UniProt P02730; UniProt P02724). In disease states, mutations in these components can lead to structural defects like hereditary spherocytosis, while pathogens such as Plasmodium falciparum exploit membrane proteins like Basigin for host cell entry (Gallagher, 2013; Crosnier et al., 2011). From a pharmacological perspective, the RBC membrane is a site for drug-induced toxicities, a target for preventing immune-mediated hemolysis, and a potential vehicle for drug delivery systems (StatPearls, 2023).
