Reduced Folate Carrier Protein 1 (RFC-1), encoded by the SLC19A1 gene, is the primary transporter responsible for the uptake of reduced folates into mammalian cells. It is essential for maintaining intracellular folate concentrations necessary for DNA synthesis, repair, and methylation. RFC-1 also mediates the uptake of antifolate drugs like methotrexate, and genetic polymorphisms can affect drug efficacy and toxicity. Recent studies indicate a role in importing immune second messengers such as cGAMP.
