Overview
Reduced Folate Carrier Type 1 (RFC1), also known as solute carrier family 19 member 1 (SLC19A1), is a ubiquitously expressed membrane transporter that plays a central role in the cellular uptake of reduced folates and antifolate drugs such as methotrexate. It is the primary route for folate entry into mammalian cells, which is essential for nucleotide synthesis and one-carbon metabolism. Loss or mutation of RFC1 can lead to profound physiological/developmental consequences due to impaired DNA synthesis and resistance to antifolate chemotherapy drugs.
