"Reduced gastric volume" refers to a physiological or anatomical state characterized by a decrease in the physical capacity of the stomach or a reduction in the volume of its secretions. Anatomical reduction is primarily a therapeutic goal in the surgical management of obesity through procedures like sleeve gastrectomy or Roux-en-Y gastric bypass, and endoscopic interventions such as intragastric balloons, which promote early satiety by limiting the quantity of food the stomach can hold [1, 11]. In a pharmacological context, reduced gastric volume describes the decrease in gastric juice and acid secretion achieved by drugs such as Histamine H2 receptor antagonists (e.g., famotidine) and proton pump inhibitors (e.g., omeprazole) used to treat acid-related disorders [6]. While not a molecular target itself, this state significantly alters gastrointestinal physiology, including the rate of gastric emptying and luminal pH, which in turn impacts the pharmacokinetics of orally administered medications [8, 12]. Clinical challenges associated with reduced gastric volume include the risk of dumping syndrome, nutrient malabsorption, and altered absorption of medications with narrow therapeutic windows [5, 9].
