Overview
Regenerating islet-derived protein 4 (REG4) is a secreted C-type lectin protein encoded by the REG4 gene on human chromosome 1. REG4 is expressed in various tissues, notably the gastrointestinal tract, pancreas, and ovary, and upregulated in several types of malignancies. It promotes cancer cell proliferation, survival, migration, and resistance to therapy through activation of EGFR/Akt and β-catenin-dependent pathways and interacts with cell surface molecules such as CD44 and G protein-coupled receptor 37. REG4 is emerging both as a promising biomarker of cancer progression and prognosis and as a potential therapeutic target. Its physiological function includes modulation of inflammation and tissue regeneration, with aberrant expression contributing to the aggressive behavior of tumors.
