Region of Interest (ROI) Brain Activity refers to the physiological and metabolic processes occurring within a specific, predefined anatomical or functional area of the brain (Poldrack, 2007). It is a fundamental concept in neuroimaging, where researchers use techniques such as functional Magnetic Resonance Imaging (fMRI) or Positron Emission Tomography (PET) to quantify changes in blood flow, oxygenation, or glucose consumption as proxies for neural firing (StatPearls, 2023). While not a molecular target itself, ROI activity serves as a critical readout for understanding how the brain processes information and how it is affected by various pathologies (NIH, 2022). In clinical research, alterations in activity within specific ROIs, such as the hippocampus or prefrontal cortex, are used as biomarkers for neurodegenerative and psychiatric conditions (NIMH, 2023). Pharmacological agents modulate ROI activity indirectly by binding to specific receptors, transporters, or enzymes located within those neural circuits. Consequently, measuring ROI activity is essential for evaluating drug efficacy and understanding the systemic impact of therapeutic interventions on brain function.
