Overview

Regnase-1 is a rapid-response ribonuclease and RNA-binding protein (RBP) that plays a central role in post-transcriptional regulation of gene expression, particularly within the immune system. It promotes mRNA decay through its endoribonuclease activity, specifically targeting stem-loop structures within the 3′ untranslated regions (UTRs) of target mRNAs. It acts as a master regulator for cellular adaptation to microenvironmental changes by restricting inflammation and maintaining immune homeostasis. Its expression is rapidly induced under conditions such as microbial infection, exposure to inflammatory cytokines, or chemical/mechanical stimulation. Dysfunction or loss of Regnase‑1 results in enhanced inflammation and autoimmune pathology, while pharmacological modulation presents therapeutic opportunities for treating inflammatory diseases and certain cancers.
