Overview

Regulation of nuclear pre-mRNA domain-containing protein 1B (RPRD1B) is a transcriptional regulator that interacts with the C-terminal domain (CTD) of RNA polymerase II, serving as a scaffold to recruit phosphatases and modulate CTD phosphorylation during transcription initiation and elongation. RPRD1B enhances the expression of cell cycle-related genes—including Cyclin D1, CDK2, CDK4, and CDK6—by promoting chromatin looping and facilitating RNA polymerase II recruitment and recycling at gene promoters and termination sites. This protein is preferentially overexpressed in a broad range of solid tumors, correlating with increased cell proliferation, tumor growth, and poor clinical outcomes. Beyond cancer, RPRD1B is implicated in the cellular response to heat shock, ensuring proper expression of heat shock proteins and survival post-stress. At the molecular level, RPRD1B forms homo- and heterodimers with RPRD1A and binds preferentially to CTD repeats phosphorylated at serine residues. While no drugs currently target RPRD1B directly, its oncogenic role and involvement in tumor cell proliferation position it as a potential therapeutic target or biomarker in cancer and stress response research.
