Overview

Regulator of chondrogenesis RNA (ROCR) is a long non-coding RNA (lncRNA) identified as crucial for the differentiation of mesenchymal stem cells (MSCs) into chondrocytes[1][2][3]. ROCR is specifically upregulated during chondrogenesis and functions by promoting the expression of SOX9, the master transcription factor required for cartilage development[1][2]. Loss of ROCR impairs chondrogenesis by reducing SOX9 induction and consequently lowering the expression of cartilage matrix genes such as COL2A1 and ACAN, along with incomplete extracellular matrix formation[1]. ROCR is located approximately 94 kb upstream of the SOX9 gene and is recognized as cartilage-restricted, exhibiting tissue-specific expression[1][2]. Unlike enzymes, receptors, or transporters, ROCR is not a traditional therapeutic target, but rather a regulatory RNA involved in cartilage biology. There are currently no drugs known to act on ROCR, no established disease biomarkers related to it, and no reported safety concerns or therapeutic challenges associated with its modulation in clinical use. ROCR may play roles in cartilage-related diseases where its expression or function could affect chondrocyte formation and cartilage maintenance[1][2][3].
