Overview

Regulator of chromosome condensation 1 (RCC1) is the only known guanine nucleotide exchange factor (GEF) for the nuclear Ras-like G protein Ran.[3][4][7][8] RCC1 is a chromatin-associated nuclear protein that binds to nucleosomes and facilitates the conversion of Ran-GDP to Ran-GTP, creating a RanGTP gradient critical for a number of cellular processes.[2][3][4] RCC1 plays a central regulatory role in cell cycle progression, especially the G1/S and G2/M transitions, mitosis, chromosome condensation, spindle assembly, nuclear envelope reformation, and nucleocytoplasmic transport.[1][2][3][4][5] It is essential for proper cell division and DNA synthesis monitoring.[1][5] Dysregulation of RCC1, including altered expression levels, has been implicated in tumorigenesis and disease states, notably via its impact on genome stability and cell proliferation.[3][5][8]
