Regulator of early neurogenesis 1 (RENO1) is a deeply conserved intergenic long noncoding RNA (lncRNA) gene required for the early stages of neuronal differentiation in mouse embryonic stem cells. Knockdown or depletion of RENO1 leads to inhibition of neuronal differentiation and massive alterations in gene expression, including failure to induce a neuronal gene expression program, reduced expression of neuronal markers, and global reduction in chromatin accessibility at key developmental regulatory regions. RENO1 operates in concert with its adjacent protein-coding gene Bahcc1, forming a regulatory circuit critical for neuronal commitment. There is currently no evidence that RENO1 is directly targeted by drugs or used as a biomarker, nor is it classified as a conventional therapeutic target.
