Regulator of G protein signaling 22 (RGS22) is a protein that acts as a GTPase-activating protein for G protein alpha subunits, promoting their inactivation and thus negatively regulating heterotrimeric G-protein-mediated signal transduction[4][5][6]. RGS22 is primarily expressed in the testis, localized to spermatogenic and Leydig cells, and is involved in spermiogenesis, particularly through interactions with GNA12, GNA13, and GNA11 G protein subunits[1][2][3][7]. Beyond the testes, RGS22 appears to have tumor suppressor activity in some epithelial cancers, where it can inhibit cell migration and invasion, likely via regulation of actin cytoskeleton dynamics through the G12/13 α subunit pathway[7]. RGS22 variants have also been implicated in susceptibility to basal cell carcinoma[7]. No approved drugs are currently known to directly target RGS22, and there is no direct evidence for its use as a clinical biomarker or established therapeutic safety concerns at this time[5][7].
